چکیده

The aminoacylation of tRNA has been fommlated as a two step reaction [ 11. The first reaction is the activation of the amino acid, and the second is the transfer of the activated amino acid onto the tRNA. Takeda and co-workers have proposed that only the first reaction requires magnesium ions; whereas, the magnesium ions are not necessary for the overall reaction [2,3]. This concept has caused confusion and necessitated a revision of the theory of the two step reaction [5]. We have reinvestigated the effects of spermidine and magnesium ions on the aminoacylation of several tRNAs according to the procedures previously reported [ 2-41. The experiments described herein lead to conclusions essentially different from those of Takeda and Igarashi [2]. Magnesium ions are essential in the aminoacylation of most, if not all tRNAs and cannot be substituted by spermidine.

Polyamine stimulation and cation requirements of rabbit liver tRNA nucleotidyltransferase.

We have examined the cation requirements of rabbit liver tRNA nucleotidyltransferase. The enzyme had an absolute requirement fo a divalent cation which could be satisfied by Mg2+, Mn2+ or Co2+. In contrast to the Escherichia coli enzyme, we have found no evidence to implicate Zn2+ in the action of rabbit liver tRNA nucleotidyltransferase. We have also identified a second cation requirement whic...

Kinetics of tRNA folding monitored by aminoacylation.

We describe a strategy for tracking Mg²⁺-initiated folding of ³²P-labeled tRNA molecules to their native structures based on the capacity for aminoacylation by the cognate aminoacyl-tRNA synthetase enzyme. The approach directly links folding to function, paralleling a common strategy used to study the folding of catalytic RNAs. Incubation of unfolded tRNA with magnesium ions, followed by the ad...

Glutamyl transfer ribonucleic acid synthetase of Escherichia coli. Study of the interactions with its substrates.

The binding of the various substrates to Escherichia coli glutamyl-tRNA synthetase has been investigated by using as experimental approaches the binding study under equilibrium conditions and the substrate-induced protection of the enzyme against its thermal inactivation. The results show that ATP and tRNAGlu bind to the free enzyme, whereas glutamate binds only to an enzyme form to which gluta...

The effect of salt solutions on the acceptance of amino acids by transfer ribonucleic acid.

The presence of dissolved salt during the enzymatic aminoacylation of transfer RNA diminishes the amount of aminoacyl-tRNA formed at the completion of the reaction. Experiments indicate that, although salt inhibits aminoacyltRNA synthetases, the diminution of the final level of aminoacylation is the result of salt-dependent changes in tRNA. tRNAs accepting different amino acids differ widely in...

Pyrophosphate mediates the effect of certain tRNA mutations on aminoacylation of yeast tRNA(Phe).

The influence of pyrophosphate hydrolysis by inorganic pyrophosphatase on homologous aminoacylation of different yeast tRNA(Phe) mutants was studied. The addition of pyrophosphatase significantly improved the aminoacylation efficiency of tRNA(Phe) structural mutants as well as the mutant with substitution at position 20, while having no effect on the charge of wild-type tRNA(Phe). Aminoacylatio...
